Pesticides applied to crops, some of which adhere to the crops, while others are scattered in the soil, atmosphere, and water environment. Some of the remaining pesticides in the environment will be absorbed by plants. Residual pesticides are directly transmitted to humans and livestock through plant fruits, water, and the atmosphere, or ultimately transmitted to humans and livestock through the environment and food chain. The main cause of excessive pesticide residues in vegetables is the prohibition of the use of organophosphate pesticides and carbamate pesticides in vegetable production by some countries, such as Methamidophos, Omethoate, Methamidophos, Parathion, Methyl Parathion, etc. Consuming food containing a large amount of highly toxic and highly toxic pesticide residues can lead to acute poisoning accidents in humans and animals. According to the national standard GB/T5009.199-2003, organophosphate or carbamate pesticides have inhibitory effects on acetylcholinesterase, thereby affecting the color reaction between acetylcholinesterase, thioacetylcholine (as substrate), and dithiodinitrobenzoic acid (as color reagent). The pesticide residue in the sample can be calculated based on the depth of the solution color after the reaction; If the degree of inhibition of acetylcholinesterase is more severe, the amount of yellow compounds generated by the reaction is also less, indicating that there are more pesticide residues in the test sample, and vice versa; The generated yellow substance has a unique absorption peak at 412 nm, which can be used for quantitative determination of organophosphorus or carbamate pesticides in the sample.
The detection results are expressed in terms of the degree of inhibition (inhibition rate) of acetylcholinesterase, and the calculation formula is: inhibition rate (%)=(Δ Ac - Δ As)/Δ Ac × 100%; In the formula, Δ Ac represents the difference in absorbance values between the control group 3 minutes later and 3 minutes ago; Δ A s is the difference between the absorbance values of the sample group after 3 minutes and before 3 minutes.

[Reagent Preparation]

③ Color reagent: Add 10ml of buffer solution to the bottle labeled as color reagent. Refrigerate and store at 0-5oC environment.
④ Enzyme reagent: Add 10ml of buffer solution to the bottle labeled as enzyme reagent. Normally, it should be stored in a refrigerated environment at 0-5oC.
When the above reagents are not in use, they should be stored refrigerated at 0-5oC; When in use, take it out of the refrigerator and place it at room temperature for testing.
[Operation Method]
1) Blank control test:

2) Sample testing:

B dried tea: Weigh 0.5g of tea sample, add 5mL of acetone, shake and extract for 5 minutes; Take 0.2mL of the above acetone solution and evaporate it in a water bath at 40-50 ℃; Add 2.5mL of buffer solution and shake for 2 minutes; Filtering, the filtrate is the sample extraction solution. (Bring your own acetone)

[Execution Standards]
